Advancements in single cell technologies have significantly deepened our understanding of the human immune system, yet challenges in sample preservation and processing have limited their accessibility, especially in resource constrained settings. The Allen Institute developed CryoSCAPE (Cryopreservation for Scalable Cellular And Proteomic Exploration) as a novel methodology that enables direct cryopreservation of whole blood without the immediate need for PBMC processing, and streamlines single cell multiomic assays.
In this webinar, Alex explores the development and validation of CryoSCAPE, highlighting its efficacy in preserving cell proportions and molecular features when compared to traditional density gradient PBMC isolation methods. He discusses the optimization of this technique for high-throughput applications, including the integration of fixed single cell RNA sequencing and liquid handling automation, and the compatibility of cryopreserved whole blood with functional assays.
