Chitinases, and chitinases-like proteins belong to Glycosyl hydrolase (GH) 18 family. Chitinases cleave chitin polymers into oligosaccharides while chitinase-like proteins (CLPs) lack enzymatic activity due to the substitution of one amino acid in their active site. The CLPs have an active-site carbohydrate binding and therefore possesses lectin-like properties. Three CLPs have been identified in mammalians, CHI3L1, CHI3L2, and CHID1. Rodents do not have the CHI3L2 ortholog. CHI3L2 is closely analogous to CHI3L1 in structure, molecular weight, nucleotides, and amino acid sequence (identity 51%). CHI3L2 was initially identified in the supernatant of primary human articular cartilage cells. CHI3L2 activates ERK1/ERK2 in human HEK293, and human glioblastoma (U87 MG). Increased expression of CHI3L2 has been associated with prognosis in breast cancer and renal carcinoma. CHI3L2 has been considered a new biomarker for gliomas, and correlates with immune infiltrates which include monocytes, tumor associated macrophages, M1 and M2 macrophages, Tregs, and exhausted T cells. Auto-antibodies against CHI3L2 has been detected in patients with rheumatoid arthritis and osteoarthritis. In addition, CHI3L1 and CHI3L2 have been associated with neurodegenerative diseases, such as Alzheimer's, multiple sclerosis, and amyotrophic lateral sclerosis.

- Human CHI3L2, amino acid sequence (Tyr27 - Leu390) (Accession: # Q15782), was expressed in CHO cells. The carboxy-terminus contains 10His-tag.
- Molecular Mass
- The 378 amino acid recombinant protein has a predicted molecular mass of approximately 42.5 kD. The DTT-reduced and non-reduced proteins migrate at approximately 40 - 42 kD by SDS-PAGE. The predicted N-terminal amino acid is Tyr.
- Purity
- > 95%, as determined by Coomassie stained SDS-PAGE.
- Formulation
- 0.22 µm filtered protein solution is in PBS, pH 7.2
- Endotoxin Level
- Less than 0.1 EU per µg of cytokine as determined by the LAL method.
- Concentration
- 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin the vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
- Activity
- Recombinant human CHI3L2 supports the adhesion of FaDu cells in a dose-dependent manner. The ED50 for this effect is 0.65 – 4.55 µg/mL.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
- Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
