Myeloid Derived Suppressor Cells and Suppression

BioLegend provides antibodies and reagents for new and intriguing cell populations, including Myeloid Derived Suppressor Cells (MDSCs). MDSCs consist of immature myeloid cells that exhibit immunosuppressive capabilities. As such, they are often recruited to tumor sites to prevent an immunological response from clearing the cancerous cells. While MDSCs are known for their continuous production of proinflammatory IL-1, IL-6, reactive oxygen species (ROS) and nitric oxide (NO), they are also known to prevent antigen presenting cell maturation and T cell activation. Lindau, et al. investigated the many suppressive mechanisms of MDSCs.

MDSCs can utilize many mechanisms to mediate suppression. CD40-CD40L interaction with Tregs induces their expansion. MDSC expression of disintegrin and metalloproteinase domain (ADAM) 17 lowers CD62L expression in T cells and immobilizes T cells. PD-L1 and PD-L2 can limit T cell responses, while TGF-b is a hallmark immunosuppressive cytokine. ROS and NO byproducts can cause CD8+ T cells to become unresponsive to peptide-loaded MHC I. Arg-1 has also been reported to downregulate TCR surface expression in T cells. Finally, MDSCs can cause NK cell anergy through TGF-b, Arg-1, or the NKp30 receptor.
