Many chronic autoimmune disorders such as systemic lupus erythematosus, psoriasis, and Sjögrens syndrome are associated with an overactive Th17 response. While ACT1 (IL-17R adaptor protein) plays a role in mediating downstream IL-17 signaling, loss of ACT1 results in spontaneous autoimmune diseases. Zhang et al. attached the DYKDDDDK epitope tag to STAT3 and the V5 or HA-epitope tag to ACT1. This allowed them to demonstrate how ACT1 directly interacts with STAT3 to inhibit downstream signaling. HeLa cells non-transfected (A) or transfected with DYKDDDDK Tag fused protein (B and C) were fixed, permeabilized, and blocked. Then the cells were intracellularly stained overnight at 4°C with 1: 250 diluted (2 μg/ml, Fig. A and B) and 1: 1000 diluted (0.5 μg/ml) Alexa Fluor® 488 anti-DYKDDDDK Tag (Fig, C, green). Nuclei were counterstained with DAPI (blue). |
