NK Cells: Editing Dendritic Cells Against Cancer

Initially discovered due to their ability to lyse multiple tumor cell lines, Natural Killer (NK) cells are key players of innate immunity. NK cells function by recognizing stressed cells that show decreased expression of markers present in normal situations e.g. MHC I. Without the need for antibodies or MHCs to prime them, these cells were aptly named natural killers. Recently, NK Cells have been found to lyse immature dendritic cells and Morandi et al. investigated this principle in vivo to determine its implications in cancer.

Perfroin, Yac-1 Cell, NK Cell, Immature Dendritic Cell, TND-a, Proinflmmatory Cell, Mature Dendritic Cell, Th1 Cell, IFN-y Cell, Tumor Cell Destruction. Yac-1 cells, which do not express of MHC I, were injected into mice to stimulate NK cells. NK cells specifically lysed immature dendritic cells (DCs) in a perforin-dependent manner. The elimination of immature DCs promoted the survival of the most immunogenic, mature DCs. These DCs helped develop a Th1 response. Mice injected with carcinomas survived at better rate when treated with Yac-1 cells, indicating NK cells could be a potentially successful therapeutic target in cancer.
