One of the most powerful tools to measure an antigen-specific T cell response is soluble MHC multimers. The T cell mediated response originates from the interaction of the T cell receptor with MHC molecules that present a peptide derived from an antigen. This TCR-peptide-MHC interaction, along with other regulatory molecules, has a major impact in the immune response. By using soluble MHC multimers, specific T cells can be identified by flow cytometry and other techniques. A key component of the reagent is the peptide loaded into the MHC, as this constitutes the specificity of the T cell. While using Flex-T™ reagents allows for custom peptides to be tested, below is a list of peptides commonly used.
