Tregs and CD4+ Foxp3+ T cells are relatively well understood in transplantation, providing immunosuppression that leads to tolerance. BioLegend provides an expansive reagent list for B cells, which are less established in these studies. Initially, the common perception was that B cells only produced antibodies that were shown to be damaging in the long-term for transplantation. Recently, regulatory B cells (Bregs) have become an interesting focal point. These Bregs have been shown to be immunosuppressive, acting mainly through IL-10 production. Chesneau et al. explored mechanisms of Breg suppression and their role in transplantation.

Bregs are thought to execute their immunosuppressive abilities through a number of mechanisms. Different Breg subsets may utilize different mechanisms. As shown in the figure adapted from Chesneau, Bregs can secrete IL-10 and TGF-?, make direct contact with cells, or inhibit antigen presentation. B cell receptor (BCR) and TLR stimulation has also been shown to induce Breg populations. These mechanisms can lead to Treg expansion (both inducible and natural Tregs), apoptosis, or inhibition of proinflammatory Th1 and Th17 cells. Chesneau's review also discusses several transplantation experiments and the role B cells played in each of them.
