    CHO cells untransfected (negative control, panel A) and transfected (positive control, panel B) with a multi-tag construct containing the V5 epitope were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes. The cells were then blocked with 5% FBS for 1 hour at room temperature. Cells were stained with 2 µg/mL of purified anti-V5 Epitope Tag (clone Poly29038), followed by incubation with Alexa Fluor® 594 goat anti-rabbit IgG (Cat. No. 406418) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 60X objective. Scale bar: 50 µm

The V5 epitope tag is used to label a protein of interest so that the tagged protein can be analyzed and visualized. Antibodies against V5 epitope tag recognize tagged proteins and allow for specific detection in several immunochemical assays such as western blotting, ELISAs and immunostaining techniques.

Polyclonal V5 antisera was raised against a synthetic peptide representing amino acid residues 95 to 108 (GKPIPNPLLGLDST) of RNA polymerase alpha subunit of simian parainfluenza virus 5 conjugated to KLH.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 0.2 µg/mL (1:5000 - 1:10000 dilution). For immunocytochemistry, a concentration range of 0.1 - 5.0 μg/mL is recommended. For immunohistochemistry, a dilution range of 1:100 - 400 is suggested.  It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is effective in immunoblotting, immunocytochemistry, and ELISA.

*Our Posi-Tag Control Protein (931301) can be used as a helpful positive control for this antibody.

The antisera was purified via affinity chromatography using the peptide immobilized on solid support.

This product may contain other non-IgG subtypes.

Product detects V5 epitope tag in ICC using Fixation Buffer (Cat. No. 420801) followed by permeabilization with either 0.5% Triton-X or 100% ice-cold methanol, or with fixation/permeabilization with 100% ice-cold methanol.
