Total cell lysates (15 µg total protein) from PC-3, A549 and HaCat cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1 µg/mL (1:500 dilution) of Purified anti-Artemis Antibody, clone 15E7C90, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. Cell lysates were loaded in order of increasing predicted DCLREC1 mRNA expression.

Description

Artemis, also known as DCLRE1C, is a nuclear protein belonging to the metallo-β-lactamase superfamily. Artemis has been found to be required for the assembling of B cell antibody genes and T cell receptor genes from individual V, D, and J segments. During V(D)J recombination, Artemis forms a complex with DNA-dependent protein kinase (DNA-‘PK) and cuts the DNA with hairpin structure. Deficiency in Artemis results in blocking of V(D)J recombination and failure to produce mature B and T cells, which is known as the condition of severe combined immune deficiency (SCID). Artemis is also found to be involved in the termini processing step of double-strand break (DSB) repair. Artemis-deficient cells are more sensitive to DSB induced by ionizing radiation or chemical agents such as bleomycin and neocarzinostatin.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody has a similar affinity to BioLegend clone 13A9C90 for human Artemis by western blot, but does not recognize mouse Artemis
