    Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-BLK (clone W21005C) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

BLK is a non-receptor tyrosine kinase primarily expressed in B lymphocytes and other cells of the B-cell lineage. BLK belongs to the Src family of kinases and plays a critical role in B cell receptor signaling, which is essential for the activation, differentiation, and proliferation of B cells. Phosphorylation of target proteins by BLK helps transmit signals that guide B cell development and activation. Dysregulation of BLK has been associated with various immune disorders, including autoimmune diseases and certain types of B cell malignancies. Outside of B cells, BLK is also expressed in a variety of immune and non-immune cells including plasmacytoid dendritic cells, pancreatic β cells, and bone marrow progenitor cells.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product (clone W21005C) may cross-react with other members of the Src kinase family.

This product (clone W21005C) is not recommended for use in immunocytochemistry (ICC) or immunohistochemistry (IHC).

For use in intracellular flow cytometry (ICFC), permeabilization with True-PhosTM Perm Buffer (Cat. No. 425401) is recommended. Fixation/permeabilization with Cyto-FastTM Fix/Perm Buffer Set (Cat. No. 426803) is not recommended.  

This antibody does not cross react with mouse BLK in Western Blot (WB).
