Total cell lysates (15 µg protein) from HT29, HCT116 and Raw264.7 treated with PMA 100ng/mL for one hour or without treatment were resolved by 4-12% Bis-tris gel electrophoresis, transferred to nitrocellulose, and probed with purified anti-c-Fos (clone poly6414). Proteins were visualized using a goat anti-rabbit IgG secondary antibody conjugated to HRP and chemiluminescence detection. Purified anti-GAPDH Antibody was used as a loading control.

    Whole cell extracts (15 µg protein) from serum-starved HeLa cells untreated (-) or treated (+) with 200 nM PMA for 4 hours were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane and probed with a 1:1000 dilution of Purified anti-c-Fos Antibody, clone Poly6414, overnight at 4°C. Proteins were visualized by chemiluminescence detection HRP anti-rabbit IgG Antibody (Cat. No. 410406) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

Description

The c-fos protein, also known as FBJ murine viral (v-fos) oncogene homolog , is a 62 kD cellular oncogene. The c-fos protein contains a bZIP region with a DNA-binding domain and a leucine zipper domain. Together with c-jun, c-jun proteins form the heterodimeric AP-1 complex that binds to the TRE elements of cellular and viral genes including collagenase, interleukin 2, stromelysin, SV40 and polyoma among others. c-fos is expressed in a variety of normal adult tissues and has been implicated as a critical transcription factor for skeletal formation and maintenance. This transcription factor is modified by phosphorylation by a number of upstream kinases and has been shown to interact with a variety of other proteins involved in transcriptional regulation. c-fos has been implicated in the development of fibrous bone dysplasia and osteosarcoma. The c-fos protein has been shown to interact with a variety of other proteins including c-jun, TFIID, NCOA2, NFAT1, CHOP, ELK1, SRC1, and ETS1.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 1:1000 - 1:4000 antibody dilution buffer for each mini-gel. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product may contain other non-IgG subtypes.

This clone is not recommended for ChIP (Chromatin Immunoprecipitation) assays (as determined by in-house testing).

Regulation
c-fos levels are highly expressed in human term fetal tissues as compared to normal human tissues. c-fos levels have been reported to be increased in patients with fibrous bone dysplasia.
