NF-kB represents a family of transcription factors that are normally inactive in the cytoplasm by binding inhibitory molecules of the IkB proteins. In response to various stimuli such as: inflammatory cytokines, bacterial or viral products, or various types of stress, the IkB proteins become phosphorylated by Iκ B kinase (IKK) complex. This leads to the polyubiquitination and destruction by the proteasome. Consequently, free NF-kB enters the nucleus and activates transcription of a variety of genes involved in immune and inflammatory response, cell adhesion, growth control, and inhibition of apoptosis. The IKK complex consists of the regulatory subunit IKKγ/NEMO (NF-κ B Essential Modifier) and two catalytic subunits (IKKα and IKKβ). IKKγ is an adaptor protein involved in activation of the IKK kinases and has been shown to be essential for the canonical and stress-induced NF-κ B pathway. Recently, Medunjanin S, et al reported that GSK-3β is critically important for ordered NF-κB signaling through modulation of IKKγ phosphorylation.
IKKγ deficiency in mice resulted in a lack of detectable NF-κ B DNA-binding activity and a lethal embryonic phenotype because of severe liver damage due to massive apoptosis. Two distinct X-linked human diseases, incontinentia pigmenti (IP) and anhidrotic ectodermal dysplasia associated with immunodeficiency (EDA-ID), have been linked to IKKγ dysfunction; indicating the critical roles of IKKγ in human development, skin homeostasis, and innate/acquired immunity.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg per ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
