Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-Reptin (RuvBL2) Antibody (clone O91A6) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular weight marker.

    HeLa cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with methanol for 10 minutes, and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with a 1.0 µg/mL (1:500 dilution) of either Purified Mouse IgG1, κ Isotype Ctrl Antibody (Cat. No. 400102, panel A) or Purified anti-Reptin (RuvBL2) Antibody (clone O91A6, panel B) overnight at 4°C, followed by incubation with Alexa Fluor® 594 Goat anti-mouse IgG Antibody (Cat. No. 405326) at 2.0 µg/mL. Nuclei were counterstained with DAPI, and the image was captured with a 60X objective.

    Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.0 µg of Purified Mouse IgG1, κ Isotype Ctrl Antibody (Cat. No. 400102) or Purified anti-Reptin (RuvBL2) Antibody (clone O91A6). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Reptin (RuvBL2) Antibody (clone O91A6). Proteins were visualized by chemiluminescence detection using HRP anti-mouse IgG, light chain specific Antibody. Lane M: Molecular weight marker.

Description

Reptin (RuvBL2) belongs to the AAA+ (ATPases associated with diverse cellular activities) superfamily of proteins and contains Walker A and B motifs, which modulate the binding and hydrolysis of ATP. Reptin (RuvBL2) plays a role in chromatin remodeling and transcriptional regulation, thereby promoting the growth and survival of cells. Chromatin remodeling complexes such as Tip60 and Ino80 require Reptin (RuvBL2) for the regulation of DNA damage responses and repair. In addition, Reptin (RuvBL2) interacts with oncogenic transcriptional factors such as c-Myc and β-catenin. Overexpression of Reptin (RuvBL2) is associated with the development and poor prognosis of hepatocellular carcinoma.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone recognizes both human and mouse Reptin (RuvBL2).

Methanol permeabilization produced stronger signal than Triton X-100 permeabilization in ICC testing.
