Western blot of purified anti-transthyretin antibody (clone A16143A). Lane 1: Molecular weight marker; Lane 2: 200 ng of human transthyretin recombinant protein; Lane 3: 4 µg of human plasma. The blot was incubated with 5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No.405306). Enhanced chemiluminescence was used as the detection system. Clone A16143A detects both TTR monomers (~16 kD) and oligomers in human plasma. -
Western blot of purified anti-transthyretin antibody (clone A16143A). Lane 1: Molecular weight marker; Lane 2: 20 µg of human liver lysate; Lane 3: 20 µg of mouse liver lysate; Lane 4: 20 µg of rat liver lysate. The blot was incubated with 5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No.405306). Enhanced chemiluminescence was used as the detection system. Clone A16143A detects both TTR monomers (~16 kD) and oligomers in liver lysates. -
IHC staining of purified anti-transthyretin antibody (clone A16143A) on formalin-fixed paraffin-embedded human liver tissue. Following antigen retrieval using sodium citrate H.I.E.R, the tissue was incubated with 5 µg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Transthyretin (TTR) is a carrier protein capable of transporting thyroid hormones in the plasma and cerebrospinal fluid, and retinol (vitamin A) in the plasma. TTR can form aberrant, insoluble amyloid fibrils that primarily deposit in a patient's heart, peripheral nerves, or both. The wild-type protein is associated with senile systemic amyloidosis (SSA) and is typically diagnosed in men over 70-years-old. Single or double mutations in TTR are primarily associated with familial amyloidotic polyneuropathy (FAP) or familial amyloid cardiopathy (FAC) which affects younger patients, often in their 30s. Clinical symptoms of TTR amyloidosis have historically been attributed to fibrils, but recent experimental evidence indicates that soluble TTR aggregates are also pathogenic.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 10 µg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 1.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
