Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1 µg/mL of Purified anti-ZNF451 (clone W19095A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. Following antigen retrieval using Tris-EDTA pH 9.0, the tissue was incubated with 5 μg/mL of Purified anti-ZNF451 (clone W19095A) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. Description

ZNF451, or Zinc finger protein 451, exhibits E3 SUMO ligase activity along with zinc finger domains that regulate biological processes. These processes include sumoylation, TGF-β signaling, and the stability of promyelocytic leukemia (PML) bodies within the nucleus. ZNF451 contains sumo-related domains which enables SUMO-protein ligase activity and interaction with SUMO 2. In mouse models, the sumoylation of PML proteins by ZNF451 decreases the stability of PML bodies. In addition, in response to cellular stressors such as topoisomerase 2 (TOP2) activity and DNA damage, ZNF451 binds to TOP2 leading to the recruitment of TDP2. This recruitment facilitates the unloading of the TOP2 cleavage complex to regulate DNA repair mechanisms. The zinc finger domains of ZNF451 effectively bind to SMAD2/3/4 proteins and attenuate upstream TGF-β signals. This interaction results in the repression of transcriptional activity, thereby influencing downstream cellular processes.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody (clone W19095A) is not suitable for immunocytochemistry (ICC).

For immunohistochemistry (IHC), the recommended antigen retrieval buffer is either Tris-EDTA pH 9.0 (10 mM Tris, 1 mM EDTA) or Citrate Buffer, 10X (Cat. No. 420902).
