Human peripheral blood lymphocytes were stimulated with 10 µg/mL of Purified F(ab')2 Goat anti-human IgG, IgM (H+L chain) antibody, clone Poly23973 (left) or unstimulated (right) for 2 minutes, fixed with Fixation buffer, permeabilized with Intracellular Staining Permeabilization Wash Buffer, then stained with CD20 APC and ZAP70 Phospho (Tyr319)/Syk (Tyr352) PE.
Poly23973 reacts with both human IgG and IgM. It can be used in functional assays, such as B cell activation. It has minimum cross-reactivity with bovine proteins. The optimal working concentration should be determined empirically based on the specific assay of interest.
0.2 µm filtered in phosphate-buffered solution, pH 7.2, containing no preservative.
Preparation
The antibody was purified from antisera by a combination of pepsin digestion and immunoaffinity chromatography using antigens coupled to agarose beads.
