Alpha-2-macroglobulin (A2M) is a large plasma protein that plays a crucial role in the immune system and homeostasis. Secreted primarily by the liver, A2M mainly functions as a broad-spectrum protease inhibitor, capable of trapping and neutralizing a wide variety of proteases. This action is protective against excessive tissue damage during inflammation and other immune responses. A2M also has roles in binding and clearing various cytokines, growth factors, and other bioactive molecules from the bloodstream, thereby regulating their activity. Finally, A2M is also involved in the clearance of protein fragments and other cellular debris, contributing to overall tissue repair and maintenance. Due to its role in protein clearance and inflammation modulation, A2M may also play a role in the progression of Alzheimer’s disease.

IHC-P, ICFC - Verified - Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

This antibody (clone A21084F) does not cross-react with mouse A2M in Western Blot.
For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
This product is not recommended for use in immunocytochemistry (ICC).
For use in intracellular flow cytometry (ICFC), fixation with fixation buffer (Cat. No. 420801) and permeabilization with True-PhosTM Perm Buffer (Cat. No. 425401) is recommended.
