Total cell lysates (15 µg protein) from PC3 (lane 1), HeLa (lane 2), HaCaT (lane 3), 293E (lane 4) and NIH3T3 cells (lane 5) were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 2 µg/mL (1:250 dilution) of Purified anti-Bcl-XS/L Antibody, clone 2H12 (upper panel). Proteins were visualized by chemiluminescence detection (Cat. No. 426303) using a 1:3000 diluted anti-mouse-IgG secondary antibody conjugated to HRP for Purified anti-Bcl-XS/L Antibody or 1:5000 diluted Direct-Blot™ HRP anti-GAPDH Antibody, clone W17079A (lower panel). Lane M: Molecular weight ladder.

Description

Bcl-X belongs to Bcl-2 family, and has several alternative splicing isoforms. It plays a critical role in control of apoptosis. The large isoform Bcl-XL (26kD) contains the highly conserved BH1/BH2 domain and protects cells from apoptosis, whereas the short isoform, Bcl-XS (19kD), which lacks of conserved BH1/BH2 domain, promotes apoptosis by interacting with Bcl-XL and Bcl-2. Bcl-XS is expressed at high levels in cells that undergo a high rate of turnover, such as developing lymphocytes. In contrast, Bcl-XL is found in long-lived, post-mitotic cells, such as adult brain.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
