Whole cell extracts (15 µg total protein per lane) from indicated cell lines and tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-CD142 (clone A22071J) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

CD142, also known as Tissue Factor (TF), Coagulation Factor III, and Thromboplastin, is a 45 kD type I transmembrane glycoprotein. It is expressed on the surface of a variety of cells that are physically separated from the circulating blood which include smooth muscle cells, fibroblasts, keratinocytes, glomerular epithelial cells (cytoplasmic inclusions), astrocytes, myocardium, liver stromal cells, pancreas cells, and is also expressed on activated monocytes and stimulated endothelial cells. CD142 is a high-affinity receptor for coagulation factor VII/VIIa and initiates the extrinsic pathway of blood coagulation. CD142 also plays an important role in a variety of diseases such as sepsis, atherosclerosis, and cancer.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody (clone A22071J) is not recommended for immunocytochemistry (ICC) or Immunohistochemistry in formalin-fixed paraffin-embedded tissue (IHC-P).
