IHC staining with Purified anti-CD4 (clone W23082E) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated either without (panel A) or with (panel B) Purified anti-CD4 (clone W23082E) (magenta) and Purified mouse anti-CD8a (clone C8/144B) (Cat. No. 372902) (yellow), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) and Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged. Scale bar: 50 µm -
Whole cell extracts (15 µg total protein per lane) from indicated cell lines and/or tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-CD4 (clone W23082E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

CD4 is a single-chain type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. It is expressed on most thymocytes, helper T cells, type II NKT cells, and monocytes/macrophages. CD4 is part of the TCR (T-Cell Receptor)/CD3 complex and binds to the β2 domain of the MHC class II molecule, and participates in TCR signal transduction. CD4 is the receptor of IL-16 and is a coreceptor for the human immunodeficiency virus (HIV) and human herpes virus 7 (HHV-7).

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704). Staining intensity may be stronger using Citrate Buffer.
This product is not recommended for use in immunocytochemistry (ICC) or intracellular flow cytometry (ICFC).
This product does not react with mouse CD4 in Western Blot (WB) or immunohistochemistry on frozen tissues (IHC-F).
