IHC staining of purified anti-Clathrin Light Chain (CLTA) antibody (clone CON.1) on formalin-fixed paraffin-embedded mouse kidney tissue. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 1 µg/ml of the primary antibody for overnight at 4°C, followed by incubation with 2.5 µg/ml of Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI, and the slides were mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 20X objective. Clathrin consists of three heavy chains (192kD). The three light chains (LCa or LCb, 25-29kD) self-assembles into a polyhedral cytoskeletal element. The self-assembly facilitates clathrin's central role in membrane sorting and selective transport within the cell.
This antibody was raised against a peptide derived from bovine clathrin light chain corresponding to the conserved region of the light chains, residues 23-44.

Store frozen (-20 or -80°C); shipped on blue ice. Aliquot upon receipt before freezing. Avoid repeated freeze/thaw cycles.
Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10 µg/ml is suggested. For immunocytochemistry, a concentration range of 1.0 - 10 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
