    Direct ELISA showing purified anti-human TIE2 (clone A19069E) bound to plate-immobilized recombinant human Tie-2-Fc chimera (carrier-free) (Cat. No. 784306). The wells were incubated with serially diluted primary antibody at room temperature for 2 hours in 1% BSA-PBS, followed by incubation with HRP Goat anti-mouse IgG (Cat. No. 405306). TMB High Sensitivity Substrate Solution (Cat. No. 421501) was used for detection. Absorbance was measured at 450 nm.

Description

CD202b, also known as Tie2 or TEK, is a 145 kD type I transmembrane protein. It is a member of the receptor tyrosine kinase (RTK) family of proteins and is expressed by endothelial cells and their progenitors, quiescent hematopoietic stem cells (HSCs), and a subpopulation of monocytes. Angiopoietin-1 (Ang-1) is an activator of CD202b that promotes, maintains, and stabilizes mature vessels and maintains HSCs in a quiescent state. Ang-2, another ligand of CD202b, is involved in postnatal angiogenesis and in antagonizing the effects of Ang-1. Tie2 binds to Ang-4 as well.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Each lot of this antibody is verified by Direct ELISA. The optimal concentration of this reagent for the specific application and the experiment design should be established by the end-user. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes
Clone A19069E partially blocks the staining of reference clone 33.1

Recombinant protein completely blocks the staining of test clone A19069E.
