Human peripheral blood lymphocytes were stimulated with Cell Activation Cocktail (without Brefeldin A) (Cat. No. 423301) for 4 hours and surface stained with anti-human CD3 (clone UCHT1) APC.     THP-1 cells, a human monocyte cell line isolated from peripheral blood of an acute monocytic leukemia patient, were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained without (panel A) and with (panel B) 5 μg/mL of the purified anti-human/mouse Galectin-9 antibody overnight at 4°C followed by goat anti-mouse IgG (clone Poly4053) Alexa Fluor® 647 for an hour at room temperature. Nuclei were counterstained with DAPI. The image was captured with a 40X objective. Scale bar: 50 μm

Description

Galectin-9 is a mammalian lectin with a molecular weight around 50 kD. It is a member of the β-galactoside-binding family. With two conserved carbohydrate recognition domains (CRDs), galectin-9 binds small β-galactosides as well as complex glycoconjugates. HAVCR2/TIM3 has been reported as one of its ligands. Galectin-9 may be retained intracellularly or transported to the cell surface where it can be cleaved to generate a soluble form. Galectin-9 is expressed by lymphocytes, dendritic cells, granulocytes, eosinophils, astrocytes, endothelial cells, fibroblasts, and thymus epithelial cells. It can be induced by cytokines in various cell types and is involved in cell aggregation, adhesion, chemotaxis, and apoptosis. Galectin-9 induces regulatory T cells and suppresses Th1 and Th17 responses.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular flow cytometry using our Cyto-Fast™ Fix/Perm Buffer Set. For flow cytometric staining, the suggested use of this reagent is  ≤ 0.25 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration of 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance in each application.

Application Notes
Clone W21101F does not block the staining of reference clone 9M1-3.
 
Recombinant protein completely blocks the staining of test clone W21101F.
