IHC staining of purified anti-JMJD6 (clone W20240B) on formalin-fixed paraffin-embedded human testis tissue. Following antigen retrieval using Tris-EDTA (10mM Tris, 1mM EDTA, 0.05% Tween-20, pH 9.0), the tissue was incubated without (panel A) or with (panel B) 5 µg/mL of primary antibody followed by incubation with Alexa Fluor® 647 goat anti-rat IgG (Cat.     A549 cells were fixed with 4% Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 1.25 µg/mL of purified anti-JMJD6 (clone W20240B), followed by incubation with Alexa Fluor® 594 goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801, panel A). Images were captured with a 60X objective. Scale bar: 50 µm

    Whole cell extracts (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1 µg/mL of purified anti-JMJD6 (clone W20240B) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

Bifunctional arginine demethylase and lysyl hydroxylase, also known as Jumonji domain-containing protein 6 (JMJD6), is a multifunctional enzyme that plays a critical role in regulating transcription and translation. One of its primary roles is the regulation of RNA splicing, where it acts as a co-factor for RNA-binding proteins that modulate alternative splicing events. Additionally, JMJD6 can also directly modify mRNA, modulating its stability, processing and translation. Previously, JMJD6 had been proposed to act as a histone demethylase; however, this function is a matter of some dispute. 

JMJD6 has been shown to regulate immune responses, cell differentiation, and embryogenesis. Overall, JMJD6 plays an important role in coordinating different cellular pathways, and its dysregulation has been linked to various human diseases, including cancer and autoimmune disorders.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 0.5 - 5.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product only weakly recognizes mouse JMJD6 in Western Blot and is not recommended for that purpose

For ICC, fixation/permeabilization with 4% PFA followed by 100% ice-cold methanol is recommended. Fixation/permeabilization with 10% ice-cold methanol or 4% PFA followed by 0.5% Triton-X is not recommended.
