Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain lysate; Lane 3: 5 ng of BSA-conjugated synthetic human MBP peptide (a.a 88-105); Lane 4: 5 ng of BSA-conjugated synthetic human MBP peptide (a.a 88-105; p-Thr98). The blot was incubated with 0.5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

Description

Myelin basic protein (MBP) is a protein involved in the myelination of nerves in the central nervous system (CNS). MBP functions to maintain the correct structure of myelin, through its interaction with the lipids in the myelin membrane. The myelin sheath is made up of MBP and lipids, and acts as an insulator to increase the velocity of axonal impulse conduction. MBP plays a central role in the demyelinating disease multiple sclerosis (MS). A hallmark of the disease is the loss of the myelin sheath surrounding nerves, thought to be induced by antibodies against MBP. Phosphorylation of MBP at Thr98 has been shown to decrease its interaction with the lipid bilayer and lead to the destabilization of the compact myelin sheath.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 0.5 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Function
