HC staining with Purified anti-Myosin heavy chain 11 (clone SM-M5) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated either without (panel A) or with (panel B) Purified anti-Myosin heavy chain 11 (clone SM-M5) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged. Scale bar: 50 µm

    Whole cell extracts (15 µg total protein per lane) from indicated human cell lines and/or tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti- Myosin heavy chain 11 (clone SM-M5) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

Myosin Heavy Chain 11, also known as MYH11, is a key protein involved in the contractile function of smooth muscle cells. This protein is a major component of the myosin II complex, which interacts with actin filaments to facilitate muscle contraction. By utilizing ATP, MYH11 enables the sliding of actin filaments, generating the mechanical force necessary for smooth muscle contraction. This function is crucial for maintaining the tone and proper functioning of various smooth muscle tissues, including those in the blood vessels, gastrointestinal tract, and respiratory system. In addition to its role in muscle contraction, MYH11 is also important for the structural integrity of smooth muscle cells. Mutations in the MYH11 gene can lead to serious medical conditions, such as familial thoracic aortic aneurysms and dissections (TAAD), where the weakening of the aortic wall poses a significant risk.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).

For Western blot (WB), this antibody has significantly higher signal when detecting mouse Myosin heavy chain 11, as compared to human. 

This antibody is not recommended for use in immunocytochemistry (ICC), or intracellular flow cytometry (ICFC) due to dim signal.
