Description

Polymeric or monomeric Ubiquitin exists free (unanchored) or covalently attached to its ligand. Ubiquitin is conjugated to its target via an isopeptide bond either as a monomer or as one of two polymeric forms. Ubiquitin monomers in one type of polymer are covalently linked via various Lys residues (polyubiquitin). The other type of ubiquitin polymer involves monomers that are linked linearly via an initiator Met (linear polyubiquitin). Polyubiquitin’s functions depend on which Lys residue is involved in cross-linking the protein. K63-linked polyubiquitin chains exhibit nonproteolytic function in vivo which includes kinase/phosphatase activation, protein trafficking, endocytosis, DNA-damage responses, as well as in signaling processes which lead to the activation of the transcription factor NF-κ-B.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 5.0 - 10 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone HWA4C4 detects Polyubiquitin chains formed by the linkage with Lys63 residue.

It does not cross-react with any other isopeptide-linked (K6, K11, K27, K29, K33, or K48) polyubiquitinylated species.
