Whole cell extracts (15 µg protein) from the indicated cell lines, HEK-293, HeLa, BeWo, HepG2, and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-PSMA6 antibody, clone O91F3, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular Weight marker.

Description

Proteasome subunit-α type-6 protein, or PSMA6, is an essential subunit of the 20S catalytic core proteasome complex. As a component of the complex, it plays a central role in the proteolytic degradation of ubiquitinated proteins, thereby, maintaining protein homeostasis by removing misfolded, damaged, or excess proteins. It is involved in multiple pathways including spermatogenesis and peptide processing for MHC class I binding, and has been proposed as a therapeutic target for lung cancer.

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.05 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

O91F3 failed ICC testing; HeLa cells were fixed with 4% paraformaldehyde and tested using two permeabilization methods (Methanol and Triton X-100). The clone failed ICC testing due to non-specific staining of cytosol.

 

IP has not been tested on clone O91F3.
