Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.5 µg/mL of purified anti-Serpin E1 (clone A16081E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining of purified anti-Serpin E1 (clone A16081E) on formalin-fixed paraffin-embedded human placenta tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween-20 (Cat. No. 925501), the tissue was incubated without (panel A) or with (panel B) 10 µg/mL of antibody overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801) and the image was captured with a 40X objective. Scale bar: 50 µm

Serpin E1 is a member of the serine proteinase inhibitors (serpin) superfamily, which is divided into two groups: inhibitory and non- inhibitory serpins. Serpin E1 is the most important physiological inhibitor of tissue-type plasminogen activator (t-PA) and urokinase-type plasminogen activator (u-PA), and therefore is a key regulator of fibrinolysis via inhibition of the activation of the broad-spectrum serine protease plasmin. Serpin E1 forms a covalent bond with both t-PA and u-PA (1:1 molar ratio). It is known that several receptors (uPAR, LRP-1A, megalin, VLDLR, and LRP-1B) bind and mediate endocytosis of plasminogen activator-Serpin E1 complexes. Serpin E1 is present in blood and the extracellular matrix in a complex with vitronectin. Elevated Serpin E1 levels have been implicated in the pathogenesis of atherosclerosis and myocardial infarction (MI). High levels of plasma Serpin E1 in patients precede MI. Abnormal levels of Serpin E1 is also associated with metabolic syndrome, obesity, obstructive nephropathy, cancer, cardiac fibrosis, pulmonary fibrosis, and bleeding thrombosis.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
