    Stability Testing for Recombinant Mouse CD300LB. Recombinant Mouse CD300LB was aliquoted in PBS at 0.2 mg/mL. One aliquot was frozen and thawed four times (4x Freeze/Thaws) and compared to the control that was kept at 4°C (control). The samples were tested for their ability to inhibit anti-CD3 induced proliferation of human activated T cells. The ED50 for this effect is 1.0 – 5.0 µg/mL.

Description

Mouse CD300LB is a member of the CD300 family of molecules that have regulatory function in the immune system. The human and mouse families have seven and nine members respectively. The human genes are localized on chromosome 17 and the mouse in the syntenic region on chromosome 11. The CD300 receptors are type I attached membrane proteins that contain one IgV-like extracellular domain having two disulfide bonds. In addition, mouse CD300LB possesses a positive amino acid (Lys) in its transmembrane domain and a short cytoplasmic tail that lacks a signaling motif. Mouse CD300LB associates with its adaptor molecules DAP10 (DNAX-activating protein-10) and DAP12 through the positive charge amino acid (Lys) in the transmembrane domain. Studies with transduced bone marrow-derived mast cells have shown that DAP12 plays a primary role in functions of mouse CD300LB, and DAP12 and DAP10 in the human receptor. CD300LB is expressed in myeloid cells and its transduction in bone marrow–derived mast cells induces cell survival, degranulation, cytokine production, and adhesion to the extracellular matrix. Mouse CD300LB is associated with LPS-induced sepsis in mice and a soluble form of CD300LB amplifies LPS-induced lethal inflammation in sepsis. Neutrophils release a soluble form of CD300LB through a proteolytic process with possible participation of MMPs and/or ADAM-TS family members. Raw 264.7 cells, but not macrophages, also release soluble CD300LB and this release is induced by LPS and it is dose dependent. LPS induces the interaction of CD300LB to TLR4 and drives lethal cytokine responses that boost septic shock.

Molecular Mass
The 379 amino acid recombinant protein has a predicted molecular mass of approximately 42.9 kD. The DTT-reduced and non-reduced protein migrate at approximately 60 kD and 120 kD respectively by SDS-PAGE. The predicted N-terminal amino acid is Ile.
Purity
>95%, as determined by Coomassie stained SDS-PAGE.
Formulation
0.22 μm filtered protein solution is in PBS pH7.2.
Endotoxin Level
Less than 0.1 EU per µg protein as determined by the LAL method.
Concentration
10 and 25 µg sizes are bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity
Recombinant mouse CD300LB inhibits the anti CD3 (Cat. No. 317325)-induced proliferation of activated human T cells; the inhibitory effect was measured using Deep Blue Cell Viability™ Kit (Cat. No. 424701). The ED50 = 1 - 5 µg/mL.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
