IL-18 binding protein (IL-18BP) binds to IL-18 with high affinity (Kd = 400 pM), neutralizes IL-18 activity and binds to IL-18 in a stoichiometric ratio. IL-18 belongs to the Interleukin-1 family and is a potent inflammatory cytokine. IL-18 is secreted by macrophages and dendritic cells and is expressed constitutively even in the absence of an inflammatory stimulus. IL-18 induces IFN-γ production, in the presence of IL-12 or IL-15, from Th1 cells. In addition, IL-18 promotes FasL-mediated cytotoxicity by NK cells. IL-18 has been associated with inflammatory and autoimmune diseases such as emphysema, metabolic syndromes, inflammatory bowel disease, and sepsis among others. IL-37 also interacts with IL-18BP at limiting concentrations of IL-18BP and at a 50- to 100-fold molar excess of IL-37. This interaction increases the inhibitory effect of IL-18BP on IL-18-induced IFN-γ in an NK cell line. IL-18BP knockout (KO) mice have a reduced abundance of splenic NK cells; also, KO mice have shown an increase in immature NK cells and a decrease in mature NK cells. The changes in NK cells are associated with a decrease in circulating plasma IL-18. IL-18 and IL-18BP are elevated in patients with eczema; also, IL-18 and IL-18BP are high in serum of patients with chronic liver disease. In addition, patients with heart failure have shown increases in IL-18 but decreases in IL-18BP levels. Currently, IL-18BP is in a clinical trial for the treatment of adult-onset Still’s disease, an inflammatory disease, which is characterized by high levels of IL-18 in plasma.

- Molecular Mass
- The 405 amino acid recombinant protein has a predicted molecular mass of approximately 45.1 kD. The DTT-reduced and non-reduced protein migrates at approximately 70 and 140 kD by SDS-PAGE respectively. The predicted N-terminal amino acid is Thr.

- >95%, as determined by Coomassie stained SDS-PAGE.

- 0.22 µm filtered protein solution is in PBS, pH 7.2.

- Less than 0.1 EU per µg cytokine as determined by the LAL method

- 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
