Description

CD275 (B7-H2, ICOSL) belongs to the B7 family of immunoregulatory molecules and is a transmembrane glycoprotein of approximately 60 kD. Mature human CD275 consists of 238 amino acid extracellular domain (ECD), two immunoglobulin-like domains, a 21 amino acid transmembrane segment, and a 25-residue cytoplasmic domain. Within the ECD, human CD275 shares 50% and 54% amino acid sequence homology with mouse and rat CD275, respectively. CD275 is expressed on antigen presenting cells such as B cells, macrophages, dendritic cells, and in monocytes. It binds to ICOS and is expressed on activated T cells, which leads to both positive and negative effects on immune responses including its own downregulation. Mouse and human CD275 exhibit cross species binding to ICOS. CD275 interaction with ICOS is costimulatory for T cell proliferation as well as the development of B cells, plasma cells, follicular helper T cells, and germinal centers. In addition, human CD275 binds to CD28 and CTLA4. The interaction of human CD275 and CD28 can co-stimulate both human mouse naïve T cells and regulatory T cells (Treg). CD275 plays a role in the development of allergic asthma by enhancing Th2 biased immune responses, limiting Th17 responses, and promoting eosinophilic infiltration into the lungs. CD275 activation by ICOS on Tregs limits pulmonary inflammation and airway hyper-responsiveness, promotes the development of inhalational tolerance, and impairs anti-tumor immunity.

Concentration
The antibody is bottled at the concentration indicated on the vial, typically between 2 mg/mL and 3 mg/mL. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C. This Ultra-LEAF™ solution contains no preservative; handle under aseptic conditions.

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding of 0.04 μg/mL Biotinylated Recombinant Human B7-H2 (ICOSL)-Fc chimera to 1 μg/mL immobilized human ICOS-Fc chimera. ND50 range: 0.01-0.07 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone W21036I completely blocks clone 9F.8A4 but does not block clone 2D3.

Function
The CD275/ICOS signal induces T cell proliferation and secretion of both Th1 and Th2 cytokines, such as IL-4 and IFN-γ, but not IL-2 (it also induces IL-5 and TNF-α production). CD275/ICOS signal may favor the induction of Th2-type responses and thus may be involved in the negative regulation of cell-mediated immune responses against tumors. CD275 is induced by TNF-α in B cells and monocytes and downregulated in dendritic cells.

However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.
