Description

CD30, also known as TNFRSF8, is a member of the tumor necrosis factor (TNF) receptor superfamily and was originally identified as a cell surface antigen Ki-1, on Hodgkin and Reed-Sternberg cells. CD30 can be cleaved by cell membrane-anchored metalloproteinases and then released from the cell as a soluble form protein. CD30 is expressed on activated T, B, and NK cells, but not on resting T, and B cells. CD30 is also highly expressed in, among others, classical Hodgkin lymphoma (HL) and anaplastic large cell lymphoma (ALCL). CD30 interacts with a number of proteins including TRAF1, TRAF2, TRAF3, TRAF5, NPM-ALK, TRAF-interacting protein, and CD30 ligand (CD153). Signaling through CD30 is thought to limit the proliferative potential of autoreactive CD8 effector T cells and protect against autoimmunity. CD30 mediates the signal transduction that leads to the activation of the nuclear factor kappaB (NF-kB) transcription factor. Coexpression of CD30 with TRAF1 augments NF-kB activation. In addition, CD30 also has been identified as a critical therapeutic target for lymphoma immunotherapy. CD30L/CD30 signaling has been implicated in the pathogenesis of several autoimmune and inflammatory conditions, such as rheumatoid arthritis (RA). Furthermore, CD30L/CD30 signaling appears to be involved in Th1 and Th2 cell responses and plays a key role in Th17 differentiation in mice.

The Ultra-LEAF™ (Low Endotoxin, Azide-Free) antibody was purified by affinity chromatography.
Concentration
The antibody is bottled at the concentration indicated on the vial, typically between 2 mg/mL and 3 mg/mL. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C. This Ultra-LEAF™ solution contains no preservative; handle under aseptic conditions.

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding of 0.8 µg/mL mouse CD30L (Cat. No. 553602) to immobilized 1 µg/mL recombinant mouse CD30/TNFRSF8. ND50 range:  0.1 - 0.8 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

No, BioLegend does not test Ultra-LEAF™ antibodies by functional assays unless otherwise indicated. Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.
