    Western blot of purified anti-β-actin antibody (clone W16197A). Lane 1: Molecular weight marker; Lane 2: 20 µg of Drosophila head lysate; Lane 3: 20 µg of Drosophila S2 (embryonic) cell lysate. The blot was incubated with 0.1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-rat IgG (Cat. No. 405405). Enhanced chemiluminescence was used as the detection system.

Description

β-actin is a ubiquitously expressed and highly conserved 42 kD cytoplasmic protein involved in cell motility. This critical cytoskeletal component can be disrupted by drugs such as cytochalasin. Because β-actin is ubiquitously expressed in all eukaryotic cells, it is frequently used as a loading control for assays involving protein detection (such as Western blotting).

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.01 - 0.1 µg/ml in human, mouse, rat and zebrafish and 0.1 - 0.5 µg/ml in Drosophila. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Reactivity to Drosophila was only verified with the purified format.
