NIH3T3 cells were non-treated (A-C) or treated with 1mM pervanadate for 30 min (D-E) and then were fixed with 4% paraformaldehyde (PFA) for 10 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained overnight at 4 degrees with 1: 5000 diluted (0.1 µg/ml, A and D), 1: 2000 diluted (0.25 µg/ml, B and E) and 1: 500 diluted (1 µg/ml, C and F) Alexa Fluor® 594 anti-Phosphotyrosine Antibody (red). Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Description

Phosphorylation is a common modification of proteins that can result in alterations in protein function, protein-protein association, cellular localization, and protein-half life. Phosphorylation can occur on threonine, serine, and tyrosine residues. The PY20 monoclonal antibody recognizes phosphorylated tyrosine residues in all species tested (human, mouse, rat, dog, chicken, and frog). The PY20 antibody has been shown to be useful for flow cytometry, immunoprecipitation, Western blotting, and immunofluorescence staining.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 0.1 - 0.25 μg/ml (1:500 - 1:2000 dilution) is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 594 has an excitation maximum of 590 nm, and a maximum emission of 617 nm.
