FVB/N mouse frozen thymus section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 30 minutes at room temperature. Then the section was stained with 10 µg/ml of purified anti-rat CD90/mouse CD90.1 (clone OX-7) overnight at 4°C, followed by 2.5 µg/mL of Alexa Fluor® 647 Goat anti-mouse IgG1 (clone RMG1-1) (red) for two hours at room temperature. Nuclei were counterstained with DAPI (green). The image was captured with a 10X objective.

IHC staining of Alexa Fluor® 647 anti-mouse IgG1 on formalin-fixed paraffin-embedded human tonsil. Following antigen retrieval using Tris-EDTA, pH 9.0, the tissue was incubated with 5 µg/mL of purified mouse anti-CD8a (Cat. No. 372902) at 4°C. This was followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG1 (clone RMG1-1) at 2 µg/mL. Nuclei were counterstained with DAPI (blue, panel A) (Cat. No. 422801) and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale: 50 µm

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. For immunohistochemistry on frozen tissue sections, a concentration range of 2.5 - 5.0 µg/mL is suggested. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 2 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
