Whole cell extracts (15 µg protein) from MCF7 (negative control), HaCaT and A431 (reduced expression controls), Daudi, Ramos, and A20 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-BIRC3 (c-IAP2) Antibody, clone W17221A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

Description

Baculoviral IAP Repeat Containing Protein 3, or BIRC3, is a member of the Inhibitor of apoptosis (IAP) family of proteins initially characterized as a component of the TNFα receptor signaling complex. BIRC3 functions as an E3 ubiquitin ligase, polyubiqiutinating target proteins such as RIP kinases, caspases, TRAF1, and BCL10. Loss of BIRC3 results in a significant impairment of TNFα-mediated NF-κB activation signaling cascades.

Application

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone has not been tested for IP.

This clone failed to test 4% PFA-fixed Ramos cells permabilized with either methanol or Triton X-100; we do not recommend using the clone for ICC.

This clone produced additional bands between ~65 and 95 kD when tested by WB. These bands were also observed with a control antibody raised with a separate epitope. These bands may represent ubiquitinated BIRC3.
