ICC staining of purified anti-CD44 antibody (clone M588) on HeLa cells. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 5 µg/ml of the primary antibody for overnight at 4°C, followed by incubation with 5 µg/ml of Alexa Fluor® 647 goat anti-Rat IgG for one hour at room temperature. Nuclei were counterstained with Hoechst, and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale bar: 50 µm

    IHC staining of anti-CD44 antibody (clone M588) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with 10.0 μg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin HRP Kit (Multi-Species, DAB, Cat. No. 929501) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 μm.

Description

CD44 is a cell-surface glycoprotein that is expressed in a large number of cell types. CD44 functions as an adhesion molecule that is important for cell migration and intercellular interactions and adhesion. CD44 transcripts undergo complex alternative splicing, which leads to a variety of functionally distinctive isoforms of this protein. Transcription of CD44 has been shows to be partially activated by Wnt/beta catenin signaling pathway, and thus is linked to tumor development.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 5.0 - 10 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
