Claudin-6 is a member of the claudin family of proteins, which consists of at least 24 members. Claudin proteins are integral membrane proteins characterized by 4 transmembrane regions and 2 extracellular loops that form tight junctions. Abnormal expression can disrupt tight junction integrity, playing an important role in the occurrence and development of cancers. Claudin-6 in particular, has been considered a therapeutic target because it is commonly expressed in tumor tissue but not in healthy adult tissue.

- Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 10 - 1 ug/mL is suggested. For immunocytochemistry, a concentration range of 10 - 1 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using PFA-Triton X-100 and Methanol. We do not recommend PFA-Methanol fixation/permeabilization.
For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer, 10X (Cat. No. 420902), or Tris-EDTA buffer.
