Whole cell extracts (15 µg protein) from E. coli DH5α cells grown to logarithmic (log) or stationary (stat) phase were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.0625 µg/mL (1:8000 dilution) of purified anti-E. coli RNA Sigma S antibody (clone 1RS1) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Ponceau S Red was used to confirm equal protein loading. Sigma transcription factor controlling the regulation of genes is required for protection against external stresses. Possibly, it affects the expression of katE (catalase HPII), xthA (endonuclease III), the appABC operon, and other genes by modifying the affinity of RNA polymerase for the promoters.
