Total cell lysates (15 µg protein) from HeLa cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 0.5 µg/mL (1:1000 dilution, lane 1), 1 µg/mL (1:500 dilution, lane 2) and 2 µg/mL (1:250 dilution, lane 3) purified anti-Elongin B antibody, clone 10C4 (upper). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-mouse-IgG secondary antibody conjugated to HRP for anti-Elongin B antibody or 1:5000 diluted Direct-Blot HRP anti-β-Actin antibody, clone 2F1-1(lower). Lane M: Molecular weight ladder.

Description

Elongin B (also known as RNA polymerase II transcription factor SIII p18 subunit and transcription elongation factor B polypeptide 2) is a member of the UbH family containing a ubiquitin-like domain. Elongin functions as a subunit of a general transcription elongation factor that increases RNA polymerase II transcription elongation past template-encoded arresting sites in the nucleus. The Elongin BC complex acts as adaptor to link Elongin A, VHL, WSB1 or SOCS1 with a module of CUL2 or CUL5 and RBX1 to form E3 ubiquitin ligases. Clone 10C4 recognizes the human elongin B protein and is useful for Western blotting.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
