Western blot of purified anti-Galectin-2 antibody (clone A17160B). Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain lysate; Lane 3: 20 µg of mouse brain lysate; Lane 4: 20 µg of rat brain lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system. -
IHC staining of purified anti-Galectin-2 antibody (clone A17160B) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using sodium citrate H.I.E.R, the tissue was incubated with 0.5 µg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm. -

Human Galectin-2 (Gal-2) is a soluble lactose-binding lectin that was initially cloned from a human hepatoma cDNA library, and designated as L-14-II (LGALS2). The galectins are a family of lectins that possess similar carbohydrate binding specificities and conserved consensus sequences. The mammalian galectins are involved in the regulation of inflammation, cell adhesion, cell proliferation, and cell death. Galectins are divided into three groups: prototype, tandem repeat a, and chimera type. Gal-2 belongs to the prototype group that includes Galectins 1, 2, 5, 7, 10, 11, 13, 14, and 15. The prototype galectins contain one carbohydrate recognition domain (CRD) and can form homodimers. Gal-2 shares 43% amino acid sequence identity with Gal-1. Studies in rat tissues and human tumor cell lines have shown that Gal-2 is expressed in gastrointestinal tract. Gal-2 induces apoptosis of lamina propria T lymphocytes, and ameliorates acute and chronic experimental colitis in mice. In addition, Gal-2 and Gal-1 induce phosphatidylserine exposure in fMLP-activated human neutrophils, but not in resting neutrophils. Galectin-1, -2, and -3 exhibit differential recognition of sialylated glycans, and Gal-2 and Gal-3 possesses higher binding than Gal-1 to fucose-containing A and B blood group antigens

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 10 µg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 0.1 - 1.0 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
