Western blot of purified anti-Gelsolin antibody (clone CPTC-Gelsolin-2). Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

IHC staining of purified anti-Gelsolin antibody (clone CPTC-Gelsolin-2) on formalin-fixed paraffin-embedded glioblastoma brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 1 µg/mL of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

ICC staining of purified anti-Gelsolin antibody (CPTC-Gelsolin-2) on Drosophila S2 cells. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 1 µg/ml of the primary antibody for overnight at 4°C, followed by incubation wit 2.5 µg/ml of Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI. The image was captured with a 60X objective. Scale bar: 10 µm

Description

After severing, gelsolin remains bound to the barbed end of the actin filament, preventing the reannealing of severed filaments. Gelsolin is crucial for the maintenance of actin homeostasis. Cells from gelsolin null mice exhibit a variety of defects, such as pronounced stress fibers formation, defective chemotaxis, and wound healing. Dysfunction of Gelsolin has been indicated in multiple diseases include cancer, infection, inflammation, cardiac injury, pulmonary disease, and neurodegeneration. Gelsolin is proteolytically cleaved and rendered inactive in Alzheimer’s disease (AD). Overexpression of gelsolin significantly reduces amyloid beta load in brains of AD transgenic mice.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 5.0 µg per ml. For formalin-fixed paraffin-embedded immunohistochemical staining, a concentration range of 1.0 - 10 µg/ml is suggested. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
