HIF2A is a transcription factor that mediates the cellular adaptive response to hypoxic conditions, orchestrating the transcription of a wide array of target genes to enable cellular survival and homeostatic maintenance in low-oxygen environments. At an organismal level, HIF2A is also involved in essential physiological processes such as erythropoiesis and angiogenesis. Under normoxic conditions, HIF2A undergoes prolyl hydroxylation which leads to its degradation by the proteasome. Oxygen starvation inhibits prolyl hydroxylation and prevents HIF2A degradation, leading to stabilization of the protein and translocation to the nucleus to alter gene expression. Dysregulation of HIF2A has been implicated in various pathological states including cancer, where it promotes tumor growth by enhancing the ability of tumor cells to survive hypoxic conditions within the tumor microenvironment.
