    Direct ELISA showing purified anti-human CD26 (clone 5-109) bound to plate-immobilized recombinant human CD26 (DPP4) (Cat. No. 764104). The wells were incubated with serially diluted primary antibody at room temperature for 2 hours in 1% BSA-PBS, followed by incubation with HRP Goat anti-mouse IgG (Cat. No. 405306). TMB High Sensitivity Substrate Solution (Cat. No. 421501) was used for detection. Absorbance was measured at 450 nm.

Description

CD26, also known as ADA-binding protein and dipeptidyl peptidase IV (DPPIV), is a 110 kD type II membrane protein. It is a member of the peptidase and ectoenzyme family. CD26 is expressed on the membrane of mature thymocytes, T lymphocytes (upregulated upon activation), B cells, NK cells, and macrophages. CD26 cleaves off N-terminal X-Pro and X-Ala dipeptides from polypeptides. It plays an integral role as a costimulatory molecule in T cell activation. CD26 may interact with extracellular matrix proteins such as fibronectin or collagen, CD45, and ADA.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. Each lot of this antibody is verified by Direct ELISA. The optimal concentration of this reagent for the specific application and the experiment design should be established by the end-user.

Application Notes

Test clone 5-109 and reference clone BA5b completely cross-block the staining on target cells.

This product is intended for sandwich-based immunoassay development on various assay platforms requiring antibody pairs. This antibody (clone 5-109) could function as a capture antibody when paired with biotin anti-human CD26 (DDP4) (Cat No. 628153) (clone 1-10) as the detection antibody and using recombinant Human DDP4 (carrier-free) protein (Cat. Nos. 764102, 764104, and 764106) as the assay standard.
