Western blot analysis of 15µg cell lysates from human PC-3, HaCaT, HeLa (Positive), Daudi (Negative) and Raw264.7 (Mouse) cells. Samples were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, probed with 1 µg/mL anti-MLKL antibody, clone W17104A, followed by incubation with horseradish peroxidase (HRP) labeled goat anti-rat secondary antibody (Cat. No. 405405, 1:3000 dilution). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904, 1:10000 dilution). Western-Ready™ ECL Substrate Kit (Cat. No. 426303) was used for developing.

MLKL has a four-helix bundle (4HB) domain at the N-terminal and a C-terminal PsKD connected via a two-helix linker. MLKL is important in the necroptotic signaling pathways. During the process, phosphorylated RIP3 (receptor-interacting protein 3) is recruited and phosphorylates MLKL which can induce the oligomerization of MLKL. Oligomerized MLKL translates to the plasma membrane, destroying the membrane integrity and inducing necroptosis. Knockdown or inhibition of MLKL prohibits necrosis. High expression of MLKL and RIP3 are reported in inflammatory bowel disease. MLKL is also involved in endosomal trafficking and extracellular vesical generation, which is necroptosis independent.
