NFIL3, also known as E4BP4, belongs to the basic leucine zipper (bZIP) family of transcription factors. NFIL3 has been shown to bind to the promoter region of the IL-3 gene; IL-3 treatment induces the expression of NFIL3. NFIL3 regulates several immunological processes including Th2 and Th17 cytokine production, Th17 cell differentiation, conventional natural killer (NK) cell differentiation, development of non-NK ILC lineages, and the response of macrophages and dendritic cells. NFIL3 has been found to prevent apoptosis of pro-B lymphocytes and is involved in IgE class switching. NFIL3-deficient mice showed impaired development of NK cells and reduced expression of transcription factors ID2, EOMES, GATA3, and TBX21.
