Total cell lysates (15 µg total protein) from A431, THP-1, and MOLT-4 (reduced expression controls), and HEL cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-PLZF Antibody, clone W18031A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:10000 dilution (lower) Lane M: Molecular Weight marker. Predicted ZBTB16 expression data was obtained from Human Protein Atlas.

Description

Promyelocytic leukaemia zinc finger protein (PLZF) is a transcription factor that belongs to the POK (POZ and Krüppel) family of transcriptional repressors. It is involved in cell cycle control, and plays an essential role in limb development, differentiation of myeloid cells, and spermatogenesis. In certain acute myeloid leukemias, PLZF exists as a translocation product with Retinoic Acid Receptor-α (PLZF-RARα) that has a dominant negative effect on hematopoietic differentiation and is capable of activating Wnt signaling by upregulating the γ-catenin promoter. PLZF can also repress transcription of TP53 and also destabilizes p53 protein through enhanced ubiquitination.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
