Whole cell lysates (15 µg) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a 1.0 µg/mL (1:500 dilution) of purified anti-RING1 antibody (clone P88C12F4) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker.

Description

RING1 is an E3 ligase that is essential for development in higher eukaryotes and plants. It is a critical component of the Polycomb-group (PcG) multiprotein PRC1-like complex, which acts to transcriptionally repress genes through post- translational modifications that block the assembly of the transcriptional pre-initiation complex. As a component of PRC1, RING1 plays a pivotal role in monoubiquitination of Histone H2A at Lysine 119, which is linked to transcriptional repression of Hox family members.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone may cross-react with RING1B/RING2.

?Due to this clone binding to additional proteins of unknown origin by western blot, it was not tested for ICC.
