Total cell lysates (15 µg total protein) from HEK293 and HeLa cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-SIK2 Antibody, clone S15G10, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular Weight marker.

Description

SIK2 (Salt inducible kinase 2) belongs to the protein kinase superfamily, CAMK Ser/Thr protein kinase family, and AMP-activated protein kinase (AMPK) subfamily, predicted molecular weight 104 kD. SIK2 is found in adipocytes and might be involved in metabolic regulation in adipose tissue. Sik2 phosphorylates ser794 of human insulin receptor substrate-1 (IRS1) in insulin-stimulated adipocytes, playing important roles in modulating the insulin-signaling cascade of adipocytes. Clone S15G10 has been shown to be useful for western blotting, immunoprecipitation, and immunoflorence staining of human SIK2 protein.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested working dilution is 1:250. It is recommended that the reagent be titrated for optimal performance for each application.
